High-Throughput Purification (HTP)
Streamline and accelerate your compound purification
Introduction
High-Throughput Purification (HTP) refers to a workflow for the rapid isolation or purification of one or more substances of interest from crude samples. To fully leverage the speed of the available technologies, HTP is typically performed in high-density formats (i.e. microtiter plates) with minimal human intervention, on a scale compatible with analytical or semi-prep chromatography.
HTP is comprised of multiple steps that can be organized into categories such as: PreQC which typically uses analytical scale LC/MS to determine the best method of isolating the compound of interest; Prep, often using preparative chromatography, for isolating the compound of interest and collecting fractions; FractionQC, an optional step to evaluate the quality of the fractions; and Final QC where the purified, concentrated sample’s final purity is determined.
HTP workflows generate significant amounts of data, all of which you must process and evaluate rapidly so you can determine the best methods to isolate and purify your sample. While data acquisition processes have been automated, data analysis has historically been performed manually, thereby bottlenecking the capacities of HTP. Seeing this challenge in the market, Virscidian developed their best-in-class software—Analytical Studio—to alleviate manual data processing and provide you with actionable intelligence that increases the throughput of your HTP workflow.
Features
Track Samples Throughout Purification
Analytical Studio provides a substance-centric workflow that allows scientists to easily track and analyze samples throughout the entire purification process, even if data is collected from multiple different instruments. All data is linked making it easy to see the history and fate of each sample and review underlying data such as the MS and TWC.
Vendor Neutral Software
Break the chains that bind you to a single vendor – Analytical Studio can read and process data files from multiple vendors, displaying these results simultaneously and giving you the flexibility to select best-in-class instruments for your experiments.
Automate Chromatography Method Selection
Analyze multiple chromatography methods for each sample and let Analytical Studio determine the best purification method, after evaluating data from all methods in near-real time, ensuring efficient purification.
Reduce False Positives
Analytical Studio's substance-driven workflow minimizes false positives in HTP data processing. It automatically calculates isotope patterns for target compounds and their potential adducts, cross-referencing them with experimental data. Only when these patterns match does Analytical Studio confirm a substance's presence, reducing both false positives and the need for manual review.
Optimize Fraction Collection for Each Compound
Analytical Studio can help you collect higher purity and higher amounts of your compounds, faster and more efficiently. Using advanced data processing and analysis capabilities, Analytical Studio analyzes your data and determines the best way to collect the fractions you need. Fraction collection settings are optimized for each sample individually, ensuring that the best thresholds and detectors are used to trigger sample collection every time.
Integration with Gravimetric and Liquid Handling Automation
Analytical Studio Fraction Mapper directly imports offline gravimetric and liquid handling data from external devices into Analytical Studio to ensure precise and reliable dissolution and replating calculations.
Automate System Suitability Experiments
Automate your system suitability assays by customizing the method and sample criteria monitored. Enable email notifications alerting administrators when exceptions occur. Track trends for retention time, intensity, and carryover to monitor for sample and instrument-related issues.
Automatically Create Experiment-Spanning Customized Reports
Effortlessly generate tailored reports in your preferred format, whether you want results reported after each sample analysis or at the end of an entire experiment. Create standardized report templates to ensure automatic report generation and consolidate data from all stages and instrumentation throughout your HTP workflow, simplifying your data management process.
