Introduction
When performing High-Throughput Experimentation (HTE), scientists run multiple experiments concurrently in well plates, performing experiments such as designing entirely new compounds and libraries, optimizing reaction conditions, running solubility screens, or monitoring reactions over time. HTE is a complex, multi-step process that includes:
- Synthetic design and layout decisions
- Material preparation
- Reactors
- Sample plating
- Data acquisition
- Results analysis
- Results reporting and storage for learning
Crucially, no one part of the process stands alone; all parts feed into and inform each other, so all parts of HTE must be informatically connected with metadata flowing seamlessly from step to step.
While hardware tools for automating HTE are available, software tools automating plate design, layout, and visualization have been lacking. Some large pharmaceutical companies have custom-built HTE software tools, and there are a few commercial software products available, however, these suffer drawbacks and only address small parts of the HTE process.
This critical gap prompted Virscidian to collaborate with a major pharmaceutical company to create Analytical Studio Experiment Builder (AS-Experiment Builder), a user-friendly, web-based software tool tailored for organic synthetic and medicinal chemists. AS-Experiment Builder simplifies HTE design and execution by tying together all the parts of the HTE process.
Its primary goal is facilitating effective reaction visualization and execution in an array format, addressing a critical market need. AS-Experiment Builder is used in conjunction with AS-Professional, which is the data processing and visualization component of Analytical Studio Software. When processing data from HTE, AS-Professional provides numerous features that streamline the data processing and decision making.

Automated and Manual Plate Design
AS-Experiment Builder gives chemists two options for designing their experiment plate layout: automatic and manual.
Automatic plate layout: This feature, exclusive to AS-Experiment Builder, accelerates experiment design. Users specify the chemicals and conditions that will be evaluated, and the software generates optimized plate layouts, eliminating concerns about chemical placement and test coverage.
Manual Plate Layout
This option offers users the utmost flexibility. Compounds can be added by weight, volume, or molar equivalents, and each well can have a different amount of each compound added. The gradient fill capability makes it easy to vary the concentration or volume of a compound across a row, column, or the entire plate.
Streamline Experiment Iterations with Templates
AS-Experiment Builder allows chemists to save and duplicate experimental templates, simplifying the process of running many iterations of a similar synthetic route. This feature enables researchers to easily transfer technology and techniques from lab to lab, and to use duplicated templates as a starting point for further synthetic optimization.
Unmatched Plate Design Flexibility
In contrast to standard 96-well plates, AS-Experiment Builder offers unprecedented plate design adaptability. Whether limited to a single row of samples or exploring intricate experiments with varying temperatures and concentrations, the software adapts effortlessly. Users can define plate configurations, ensuring maximum flexibility in adjusting reaction conditions while benefiting from automatic experiment design.
Seamless Chemical Database Integration
AS-Experiment Builder effortlessly integrates with your organization's internal chemical databases, simplifying experimental design and ensuring the availability of required chemicals for synthesis. Moreover, the software enables links to commercially accessible databases, broadening the scope of your experiments.
Streamlined Compound ID and Reaction Yield Calculations
Within AS-Experiment Builder, users can manually create or import sets of reactions for their experiments, whether spanning a single well, an entire row or column, or even the entire plate. These reaction schemes include data like predicted reaction products, potential byproducts, and impurities. Integrated seamlessly with Analytical Studio, this compound data aids in identifying LC/MS data peaks, simplifying and automating the determination of optimal reaction conditions. Detailed compound identification information is stored in Analytical Studio, becoming an integral part of the metadata for each sample and data file.
Effortlessly Create Sample Preparation Instructions
Following the experiment's design, AS-Experiment Builder enables the automatic generation of comprehensive sample preparation instructions. These instructions simplify the creation of required stock solutions, offering detailed steps for equivalence, concentration, volume calculations, and any additional sample preparation requirements. Moreover, when employing robotics for sample preparation, these lists seamlessly transfer to the sample prep robot for streamlined execution.
Simplify Results Visualization
AS-Professional takes full advantage of the metadata transferred from AS-Experiment Builder to simplify data analysis. The results displayed in the well-plate view allow for a quick assessment of which experiments were successful, indicated here by the green coloring for wells where the target product was synthesized. When more granularity on how much product was synthesized is needed, you can view the well-plate based on percent conversion, with the brighter green indicating a high percent conversion and the yellow/orange/red indicated less conversion. For each given well, you can view the TWC which shows all the compounds found in that well, again color coded by substance, simplifying data review.
